PFAS in Drug Active Ingredient Synthesis
Fluorine chemistry is extensively used in pharmaceutical active pharmaceutical ingredient (API) synthesis, with approximately 20% of drugs containing fluorine atoms. While organofluorine drug compounds differ from industrial PFAS, some synthesis intermediates and manufacturing byproducts are PFAS compounds of regulatory concern.
Environmental Assessment Requirements
FDA’s environmental assessment requirements for new drug applications include evaluation of manufacturing process chemical discharges. PFAS-containing process wastes from fluorine chemistry drug synthesis must be characterized and managed under RCRA and Clean Water Act requirements.
Green Chemistry Approaches
Pharmaceutical green chemistry initiatives are developing PFAS-free synthesis routes for fluorinated APIs. Enzymatic fluorination, electrochemical methods, and alternative synthesis strategies reduce reliance on PFAS-generating reagents in drug manufacturing.
EMA Environmental Risk Assessment
The European Medicines Agency’s environmental risk assessment (ERA) requirements for drug approval include evaluation of manufacturing process environmental impacts. PFAS manufacturing discharges are increasingly scrutinized in ERA submissions for fluorine-containing pharmaceutical products.
